N2 - Statement of Purpose: Oxygen is required to support viability and functionality of cells in tissue engineering. However, limited oxygen supply through diffusion is a major challenge for engineering large tissue constructs, particularly those containing metabolically active cells. Embedding oxygen releasing materials in engineered scaffolds to maintain a high level of oxygen in the inner parts of large tissue constructs is one approach to addressing this challenge. Here we develop oxygen-generating microparticles by encapsulating calcium peroxide (CaO2) in polycaprolactone (PCL) through homogenization and electrospraying to sustain prolonged oxygen release. The degradation of biodegradable PCL allows the controlled release of encapsulated CaO2, which then reacts with water to generate O2 to supply a tissue construct.
