Nano-LC MALDI-QTOF-MS for the analysis of complex ICAT labeled protein mixtures

Abstract

The use of nano-LC MALDI-QTOF-MS to identify and quantify ICAT labeled protein mixtures was analyzed. Proteins expressed by a cell under two different physiological conditions were labeled with ICAT reagent on their crysteinyl residues. After washing, the labeled peptides were eluted from the avidin column and separated by reversed phase nano-LC. Results showed that the technique has great potential for bypassing the 2D gels and the study of protein expression levels from different cell states.
