Effects of TGF-β1 on cells growth and expression of plasminogen activators and its inhibitor on cultured rat mesangial cells

Abstract

Purpose: To study the effects of transforming growth factor-β1 (TGF-β1) on cells growth and expression of plasminogen activators and its inhibitor on cultured rat mesangial cells transfected with TGF-β1 gene and antisense TGF-β1 gene. Methods: MTT array, cell counting and FACS analysis for examining mesangial cells proliferation. Northern blot and Western blot analysis for detecting the levels of plasminogen activator inhibitor-1 (PAI-1) mRNA and protein, respectively. Zymography analysis for determining the activity of tissue type plasminogen activators (tPA). Results: Overexpression of endogenous TGF-β1 inhibited mesangial cell growth, increased the level of PAI-1 mRNA and protein, and decreased the astivity of tPA. Conversely, lower-expression of TGF-β1 stimulated mesangial cells proliferation, decreased the lerel of PAI-1 mRNA and protein, and increased the activity of tPA. Conclusions: TGF-β1 as a potent inhibitor of mesangial cells growth and a regulator of tPA/PAI-1 may play an important role in the development of glomerular fibrosis.
