Detection of circular DNA from mitochondria of Neurospora crassa

Abstract

Mitochondria from a slime mutant of Neurospora crassa were prepared by standard procedures and DNA was isolated by the cesium chloride-ethidium bromide technique. Electron microscope examination of the DNA banding at a position expected for closed circular DNA detected a population of circular DNA with a contour length of 20 ± 1.6 microns.
