N2 - A method for measuring ribose 1-phosphate in cell extracts is described. Cell extracts are first fractionated on polyethyleneimine-impregnated cellulose columns to remove nucleoside and base components which otherwise interfere with the enzymatic assay. Ribose 1-phosphate in the eluate is made limiting for the conversion of [14C]hypoxanthine to [14C]inosine in the presence of purine nucleoside phosphorylase. Labeled substrate and product are then easily separated on boronate gel columns or by paper chromatography.
