All-trans retinoic acid upregulates the expression of uPA/MMP-2 in cultured rat mesangial cells

Abstract

Purpose: To study the effects of all-trans retinoic acid on the expression of uPA/MMP-2 in cultured rat mesangial cells, and to elucidate the mechanism by which all-trans retinoic acid prevents age-related progressive glomemular sclerosis. Methods: The mRNA, protein and enzymatic activity of uPA/MMP-2 were detected by Northern blot analysis, Western blot analysis and Zymography assay, respectively. Results: All-trans retinoic acid upregulated the expression of uPA/MMP-2 protein, enhanced the enzymatic activity of uPA/MMP-2. Thus all-trans retinoic acid elevated the level of MMP-2 mRNA in cultured mesangial cells. Conclusions: All-trans retinoic acid upregulated the expression of uPA/MMP-2 in cultured rat mesangial cells. It is valuable for explaining the role of all-trans retinoic acid in preventing age-related glomerulsclerosis.
