Abstract
We describe a method for labeling tumor boundaries using an enzyme active in the tumor boundary to crosslink a fluorescent-labeled substrate into the boundary. Preliminary in vitro assays are described.

Funding Information:We are studying a method for specific labeling of tumor boundaries using infrared fluorescence. This method is based on the action of the enzyme tissue transglutaminase, an enzyme is involved in the wound-healing response [1], where it helps stabilize the extracellular matrix. The enzyme has also been shown to be involved in neovasculature and extracellular matrix at the boundary between tumors and normal tissue [2,3]. We plan to use the tissue transglutaminase activity to crosslink substrates conjugated to fluorescent infrared dyes to label the tumor boundary. Fig. 1 shows results of an in vitro assay for tissue transglutaminase activity based on scratch injuries of confluent EA-hy296 cells.
